Transmission-through-dye (TTD) is a technique developed at Kent State University that is complementary to bright-field microscopy. It works by adding a nontoxic blue dye to a sample and shining a red light on it at a wavelength that is strongly absorbed by the dye. Live cells exclude the dye and generate contrast in TTD images, with thicker cells or their parts appearing brighter. Broken cells and debris do not exclude the dye and remain dark, so it is easier to differentiate between living and dead matter. The TTD microscope also functions as a regular microscope, allowing you to switch between red and standard white illumination.
